There is a big difference.  $$$$$$$$ They have insurance premiums to
pay, and you don't.  Some will want to do Nitrox 1, or Nitrox 2 only.
You, as master of your own demain, can pump anything you like.  You
will also have better control over the end result.  They may tell you
that it's the percentage you asked for, but unless you have your own
analyzer.  Well, your a very trusting sole to dive a mix without
checking it yourself.  If you have your own analyzer, then why pay
someone else.
The big question is. Are you responsible enough to keep everything
clean, and in tip top shape?  Remember your life depends on it. 
If not, pay the store.  Then  if you  die,  your family will have someone
to sue.  Come to think of it.  If you don't possess the discipline to handle
this yourself.  Why even consider diving, let alone moving to other
than air mixes.
As for the quality of O2.  I beg to differ with Ronnie.  All O2
cylinders have a vacuum pulled when returned to the store.  If
purchased from Air Products anyway.  It's the number of vacuums
pulled, that differs between the various bottles.  
Air Products, by the way, has donated thousands of dollars of gas to
various projects over the years.  So if at all possible, please buy
from a respected friend of the diving community.
